Screening and Expression Analysis of ADSSL1 Gene in Jingyuan Chicken Based on RNA-seq

Abstract

The differential genes affecting meat quality and taste were screened by RNA-seq, and the expression of genes related to meat taste in breast muscle and leg muscle of Jingyuan chicken were verified by RT-qPCR. The results showed that there were 666 genes significantly differentially expressed between breast muscle and leg muscle of Jingyuan chicken, of which 256 genes were significantly up-regulated and 410 genes were significantly down-regulated (P<0.05). GO functional annotation and KEGG pathway analysis showed that adenylosuccinate synthetase isozyme 1 (ADSSL1), a differentially expressed gene related to inosinic acid, was screened from purine metabolism, alanine, aspartic acid and glutamate metabolism pathways, and the mRNA expression of ADSSL1 in chest muscle was significantly higher than that in leg muscle (P<0.01). The results of this study provide a scientific basis for the screening of inosinic acid-related candidate genes, and provide a theoretical reference for further under- standing of ADSSL1 genes.
